Research Title : Characterization of 2,3 – Dihydroxybiphenyl Dioxygenase From Hydrocarbon Degrading Bacteria

Research synopsis :

2,3 – Dihydroxybiphenyl dioxygenase (DHBP), an enzyme involved for first aromatic rings cleavage in biphenyl and polychlorinated biphenyl biodegradation pathway. This enzyme has been expressed by biphenyl degrading bacteria such as Pseudomonas pseudoalcaligenes KF707, Pseudomonas aeruginosa PAO1161 and Rhodococcus rhodochrous K37 from previous studies. We interested to study the characteristic of DHBP enzyme in our own culture collection for hydrocarbon degrading bacteria by cloning the bphC gene into cloning vector such as TOPO. The enzyme then have been expressed using Escherichia coli BL21 and the enzyme were characterized by method of purification, western blotting, enzyme assay and SDS- PAGE.
